Pharmacokinetics
Absorption: Oral bioavailability is highly variable (averaging 5% to 50%) due to first-pass metabolism. Food, particularly dairy, decreases its absorption.
Distribution: It distributes widely into body water but enters the cerebrospinal fluid (CSF) negligibly. Plasma protein binding is approximately 19%.
Metabolism: It is inactivated primarily by two pathways: thiol methylation (via the TPMT enzyme) and oxidation (via xanthine oxidase).
Excretion: It is rapidly eliminated, with a plasma half-life of less than 2 hours. About 46% of a dose is excreted in the urine within 24 hours
